Here's a thought, Astartes who are able to retain the ability to breed will be able to allow her children (should they chose to) to become Astartes without the huge chance of the gene-seed rejection.

I.E those who are able to become one will have a larger chance to be one should there mother is already a Astartes.

If they chose not to be one they'll be one hell of a elite guardsmen.

Biomancy is a thing in a legion full of psykers. If they got curious they could change that on a whim assuming Seras did the whole chemical sterilization brainwashing part of the process instead of mentally guiding this first batch throught the familiarization procedures for their new strength/size/organs. They also already had a minor astartes package so they might already be used to having these organs if not their new sizes or things like the mucranoid or betchers gland.
In short I expect some number of Astartes maybe 10 out of the 250 pyskers stated to breed true in the future.

Other ideas/tactics we can use:
-stick long range teleport beacons in artillery shells, use them to flank unexpectedly.
-Stick teleport beacons in drop pods so we can easily reinforce beachheads created by our legion.
-create dedicated mobile void shield generators
-max out ion shield research to make everything even tougher
-max out modularity research for more customizable loadouts
-do we have melta tech? If so, max it
-max out our training simulators: not only is fighting at full effectiveness on deathworlds awesome, we could be able to get bonuses against the Ork ecosystem
-have we maxed out voidcraft engines? if not, we need to, as speed lets us keep Orks out of their preferred range
-max out fixed void fortifications
-research ramilies starfort equivalent
-research teleport jammers to deny the Orks one of their favorite tactics
-research subspace as much as we can to figure out what we can use it for
 
Other stuff to do:
-create a combat doctrine and chart of organization for our legion
-use integration actions to create a logistics train for the legion (make sure they are fully supplied with initiates and war materiel)
-I like the idea of Serras making wargear for all her captains and champions. Maybe have her make staves for Librarians when they graduate from being intiates?
-expanding the legion needs to be a top priority
-we just finished improving our ship sensors, can we do the same for auspex and similar?
-put teleport beacons in our Land Raiders.
-Research miniaturization so we can stick more goodies on our stuff

We should work on the crystal weaponry for a bit after we upgrade all elemental runes, I feel they would be really effective on warp creatures. Plus gives us more obvious combat spells (those are still a thing right? we active the powers of the rune without having a rune if we can make one?)

Edit: work on 'magic' ammunition as well because our initial attempts to create it was crap.

How many words do you need for an omake?

Also, I just realized that we should have gotten bolter tech when we got the Land Raider STC. We should improve that as much as possible, and research explosive compounds as well so conventional artillery doesn't lag behind laser/plasma direct fire.

Oh, we could teleport nukes into enemy ships! We could have a fighter/bomber designed around that role...

@ilbgar123 a couple questions for you: how do our laser/plasma weapons compare to regular 40k's? How do they compare to bolters? What kind of weapon mix is our Legion using currently? Do our vehicles use promethium, or fusion tech? And finally, how safe is our current infantry-scale plasma tech? Vehicle mounted ones don't overheat, right?
 
Somewhat better lasers. Significantly better plasma. Bolters are a bit behind. 60% laser. 20% plasma. 20% mass-launcher. Fusion tech. Pretty safe, as long as it's not pointed at yourself. You can only make weapons so safe. Not unless you're in an active volcano or something.

We're researching ground tech next turn right?
I think that's the current plan. Aside from development of our Exterminatus Class Capacitor Missiles, shipyards and our planet killing super weapon it should be mostly ground tech.

Also just asking. Would it be useful for us to upgrade and build more Torus Platforms now? We're cutting it close on the 3 turn limit but if we do so we can get a potentially substantial boost in ship yards and increase our production just before the Ork Armada arrives.

Torus Platforms System. A set of space elevators connected by platforms located in low-orbit, which can be used to increase the ease with which goods and people flow across the planet and to and from it thanks to ease of movement to and from the surface from space. Bonus Modified due to Arcology Technology. Upon construction, gain 5 effective planets. A +10000 to industry. +1000 to defensive rolls for that planet and +5 of each type of shipyard. May sacrifice two of these bonuses to craft a ring with triple the bonus in the remaining category, each of the three preceding sentences being a category in this case. 2 year construction initially as space elevators occupy first year. Additional rings require 1 year.

Can the first ring be specialized? Because if we can we could get +15 of each shipyard, per ring right before the Orks arrive.

Also everyone remember the wording "each type of shipyard" which means that if we get another ship class size we can get up to 60 shipyards in total.

Thanks! Do we have some kind of plasma grenade for anti-armor work, or are we reliant on krak grenades?

All of the rings can be specialized. You don't actually need any generalist rings, though keep in mind it's 4 per planet.

Thread posters please upgrade and make more Torus Platforms. Right now if we specialize the rings for ship building, with 3 classes of ship currently that would mean 45 ship yards per ring. And that's the base level Torus Platforms System.
