So, pursuing an entirely separate path to Enlightenment is generally pointless - you would have to start all over again in the best case, and you might even need to unlearn whole parts of your original path when they conflicted with the wisdom of the new one.
However, it is entirely possible to synthesise a new path by combining two or more existing schools into a single unitary Path. The more common elements they share, the easier this is to accomplish - Vandire himself is clearly in the process of formalising a unitary Path that combines the Administratum Path (which focuses on extracting a 'tithe' from other people or the world and using it as you desire) and the Ecclesiarchal Path (which harnesses the burning zealotry of faith, yours and others). Or it would be more accurate to say that he has already done this, and is now trying to force the ascendency of his new way over its predecessors.
The Moirae believe in attempting something similar, combining the Mechanicus pursuit of Akasha with the Ecclesiarchy's use of Miracles. You're not sure if they have succeeded, but as a supporter of theirs you are certainly free to make the attempt yourself.
Independently of any such synthesis, it is also possible to learn the individual Arts and Techniques of a rival path, but this generally requires figuring out a way to adapt them to your own paradigm. Having passed through the Runic Realm, the Young Mistress is well suited to such adaptability of thought and spirit - now she just needs to find teachers.

Nobody knows. For that matter nobody is entirely sure whether they are even a Sect in the traditional sense - they certainly don't seem to recruit or seek enlightenment in the traditional fashion. It is assumed that they pursue some manner of path set out for them by the Golden Emperor, but he was always secretive and capable of doing tremendous harm to anyone who tried to steal his wisdom, so the specifics are not well known.
This whole post was great if course, but I love this line especially. It really sells the vastness of 40K that you can't tell if you are underground or in a ship.

The issue is that menials are fragile, and being hauled bodily through the air in a hurry (and by a woman who cannot use her hands to provide additional control if she wishes to carry them) to be dumped on the ground could well cause them injury.
