Lucius, with the death of Caesar, you (surprisingly) remain standing. While you have the Legions respect (or at least the Guards'), that translates to little responsibility outside the Guard. What is being claimed: Lucius
Canonical/Original Content explanation: Lucius and his Praetorian Guard hold great responsibility and power in the vacuum of Caesar and Lanius' death. Protecting the leadership of the Legion, Lucius has his hand, and sword, close by those who matter, a fact which cannot be overlooked. Likewise, Lucius represents the ideologically primitive aspect of what Caesar stood for; the Praetorian Guard represent the professional character expected out of every legionary - obedient and skilled in a life that far preceded the apocalypse.
Why they are relevant: In command of the elite Praetorian Guard, Lucius and his subordinates possess a core fighting force, but perhaps more importantly, were tasked with protecting the leadership of the Legion. Thus, when Caesar lived and fell, the Praetorian's had every man of importance within their grasp. Likewise, Lucius stood as one of the most respected figure in the Legion after the fall of Caesar, not feared like the monstrous Lanius, but truly seen as a stoic warrior.

Canonical/Original Content explanation: The original right hand man of Caesar and a vital part of the creation of the Legion the brutal first Legate was punished for his failure against the NCR by being set ablaze. Surviving the incident the Burning Man now leads the Dead Horses in Zion.
Why they are relevant: Caesar while he was still alive deemed it worthy of hunting down the Burned Man going so far as to offer the White Legs the chance of joining the Legion to kill him. While outgunned by the White Legs there was a reason that Caesar made this effort to hunt Joshua Graham.
In a world with the Legion, the Khans look a hell of a lot better than they did in the original two Fallout games. They may be looting rapists who burn down the world and take what they want, but they have gender equality and a pretty libertarian/anarchist social structure.

Canonical/Original Content explanation: The Think Tank was the center of the brightest minds of Pre War America, and responsible for countless advancements before the bombs fell. None the less even then the pursuit of SCIENCE!!! never ended, however, infighting, and damages have led to the death of the Think Tank without the Couriers Intervention as Dr. Mobius eliminated his former colleagues and even salvaged there very brains to extract their knowledge as his own, using Big MT's advanced technology and his expertise.
This, in turn, has led to even more drastic a change in Dr. Mobius personality which settled down and took hold as he regretted his actions deciding to start anew and made plans to improve himself undoing countless damages to his body and changed mind and the decay that had been occurring on Big MT itself as he prepared himself for the beginning....of an entirely new life and with it a new name.

(Essentially without the Courier Mobius spiraled further into insanity and wiped out the think tank, regretted this too and decided to start anew, to start fresh and essentially killed himself in an operation that combined the brilliant minds of the Think Tank together into a new body, unrestricted as before, as a new person, into the unforgiving post-war world.)
Why they are relevant: A Variety of unreleased and experimental Pre-War discovery's as well as Post War ones as the Think Tank continued its experimentation over the centuries since the great war. This added on to local sources of raw material, changeable material, and working industry makes the Big MT a new awakening technological faction to be reckoned with especially with an army of a variety of robotic creations and experiments at Dr. Mobius's or rather DR PHOENIX's beck and call in between New Vegas and the NCR
