It depends on poiosn. Poison made of fresh water affect all living things... Maybe it doesn't stand on Mother Mushroom, but on some other big one in the center of the island? Also, is this Mushroom big? Because maybe there would be not enough poison or would be pouring out of the cup not fast enough to kill it?
Size doesn't matter when it comes to poison. For example, a drop of nicotine kills a horse. Plus, even if the poison does not kill the mushroom immediately, it will accumulate in it until it actually kills it. I just hope that my mushroom people will find this death machine in time and isolate it, or we will see the first extinction in the quest, which is also not bad

Im going to step in to say that poison absolutely depends on size.

Dose is the difference between many medicines and poisons.

For instance, caffeine is an insecticide.

This is not true. The dose makes the poison. People need to smoke for all thier life to finally die, because of effects of poison. And as I said, not all poisons from this cup can work infinetly long. Fresh Water poison cannot - it would affect things, but not infinitely. At some point it will leave the organism or just lose its properties. This not even counting mechanism of how specific poison works. So there are chances of survival.​

It's like putting a cyanide laced candy in a bowl of M&M's and saying there is a chance eating from it won't kill you.
Well, if the Mushroom is big enough, it would be just crippled. Probably ill, because of poison slowly seeping from the Cup. However, it is only if something or someone will not disturb the Goblet. Maybe some strong wind or wild animal could make it fall to ther Earth, even getting upside down in the process.​

Edit: I just wasn't tagreting for killing the whole island. Maybe I should change the location of the Goblet?

So, you want to make the Builders then? That could be interesting. It wouldn't be that out of place, especially if we're going to make the Second Pyramid more metal. Although it might not seem like it'd fit on the surface, if you remove a lot of the cartoonyness the Builders become very scary people.

Speaking of, I'm thinking I'm going to make my second island artifact a single weapon. Not sure what it will be, probably a sword (I'm a sucker for the classics) or a hammer. Same pattern as before, starts mundane, gets upgraded each turn. Something like, normal, then unbreakable, then runes that empower it, give the wielder a destiny or something, maybe more runes, and lastly a either a negative or some restrictions.

That's what I'm thinking right now anyways. We'll see how it goes.
 
For now I only have list of things for them. Tried to base thier characteristics on the game mechanics and a little drop of creativity.
Powers:
  1. Force - race of builders able to charge their actions with the unique Force they possess. It makes tools more efficient, weapons more dangerous, etc. It has a lot of applications - some obvious, others need to be looked for.
  2. Inventory - race of builders possess 'pocket dimension', which is able to store all items and material they need. How much depends on three things: volume, mass and diversity of materials in storage. Things inside inventory are frozen in their state, complex enough creatures can easily resist it - so it is impossible to store something with too complex mind and/or powerful will.
  3. Crafting - race of builders can change materials inside their inventory. They can do it stealthy or via projecting what they are doing in the outside world, probably letting other builders help with the project. Can be combined with Force to change things outside inventory, extend crafting ability. They get blueprints of everything getting into their inventory. So it is possible for them to create copies of objects, if they have materials needed and some tools.
  4. Building - race of builders can create things, which somewhat define conventional architecture. Like creating floating islands via making something about mass of matter. It can be broken with enough force applied.
  5. Peak Body - their Force constantly preserves their body. They are biologically immortal, heal really fast and can even regenerate lost body parts over time. Although, this still drains their bodies of material - makes them hungry and thirsty, if wounds are too severe. This also makes them much tougher, stronger and more resilient than the average human. Their bodies strive to return to their peak state, if the materials are available
  6. Awareness - they are very aware of their own bodies. It makes them more agile, dexterous and fast, and lets them learn or relearn skills much more quickly. They know if they are ill or poisoned. And if have enough experience and data accumulated through putting things into inventory, they can even pinpoint the cause of the affliction.
Complications:
  1. Isolation - they tend to be more territorial than humans, but need less social interaction and contacts to be healthy. But they tend to have small groups of very close people. They prefer to live in small communities, or even completely alone in their own created place.
  2. Children - women of this race can give birth only once in the ten years, which is connected with the soul and Force they possess. To get pregnant both man and woman need to be willing to do so - make their Forces interact in very intimate matters. Otherwise it would not work.
  3. Growing Up - children have Force from birth, but first five-six years it is passive and internal: fast healing, durability and strength, improved ability to learn motor skills. Until ten years they slowly get awareness and ability to use their Force exteriorly, they get growing inventory and other things. By the age of twelve they usually have all the same powers as an adult member of the species, but mostly lack knowledge, experience and skills.
