Maybe. But that's an overnight flight at best. Better get planning, those things usually have sun at one end of them. There's also the fact that you a flying towards the dawn. Also the vampire holy weakness will not play nice with the fact that it is the VATICAN, home of the Holy See. IF we want to be really mean, set a time limit that makes them do it sometime in the summer. When days are long and nights are short.

im thinking that since most vampires are creatures of the night with extreme paranoia and a healthy fear of sunlight a test where you have to, without any special abilities or magic, get someone to fully trust you, and that you have to Genuinely trust them back

Perhaps, but how long would it take for a centuries old vampire to confess all of their sins? Would they do so without the person hearing their sins repelling them in some way for being an undead monster? Oh yeah, there's also how younger vampires are vulnerable to holy symbols.

I don't have a better idea to make a centuries-old schemer vulnerable. I would propose adopting a baby and raising it well to adulthood, but that's just begging for unusual interpretation of "well".
 
I'll be honest @Magoose, a big problem is that this is far too nebulous a request for the questors to handle when we still know so little about the setting. There's simply not enough context to make an informed choice, so we're stuck with broad strokes and vague assumptions.

Yeah but the thing is I lost the options that I made for this months ago and…

Well I just decided to make it a write in and fill in the blanks next update for the challenge

It be really funny if, due to some old testament bible antics non of the enslaving was a sin
 
I may have an idea: a version of a bar marrathon where you need to go to 7 holy site in rome, sincerely pray an ave maria an a pater noster and finish by the full confession and pardon at saint pierre. the contestant and their allies would be banned from interfering in any way with the concurent .
