A month after Pearl Harbor, the Empire of the Rising Sun replaces Imperial Japan and all of its assets. The Empire sees this time travel as destiny giving it the ability to conquer and rule the world. How well will the Empire of the Rising Sun suceed in this task? And if they defeat all the major powers, how will the world look under Imperial tutelage? Will a man like B.J. Blazkowicz head to the far east this time and start a large uprising against the Empire?

Probably they beat America and we get a Japanese century while they are the super power the wheel of human history keeps moving America and it's struggles aren't that important.
 
There is literally nothing that the 1940s can do to offer more than a speed bump to an Empire that can churn out hundreds of plasma cannon toting battleships and thousands of six story mecha who can eat killsat blasts to the face and saw tanks in half with their eye beams. Not even getting into their mass usage of supersonic jets, guided missiles, directed energy weapons, power armor, transformers, nanotechnology, and literal combat psionics. Not to mention being able to construct mobile military cities that can house and supply massive numbers of soldiers, warships, and all their equipment to allow for extremely long-range offensive operations like invasions of the mainland of Europe and North America.

The Japanese conquer the world in weeks.

Even if the world could get out an atomic bomb fast enough before the Japanese Empire overruns all opposition (they can't), the fact that it has to be delivered by slow-moving prop-engined bombers means it'd basically be useless anyway.

How will Germany and Italy react to this super Japan coming out of nowhere and crushing America and the Soviets with ease?

And how will Japanese culture change with it being the premier superpower?

Yoshiro's goal was to conquer the world so they'd react by getting steamrolled too.
